I'm loading my beloved preset in optimaster , then i run the wizard.
Now I'm wondering if I loose part of my loaded settings by running the wizard.

sure you do.
the wizard provides a number of choices.
personally, i adjust the compressor band outputs to match the inputs(as usual for a compressor) after running the wizard. it's fast and effective. there is no need for presets...i find that the first few choices are usually closest to correct, but ymmv.

I use optimaster just for the complete mix, at the very end.
The preset-names suggest it might be an idea to have several optimasters in parallel for different tracks of the song. (like voices and piano..)
Does this make any sense ?

no, not to me. 
however, what works for you is important.
i use optimaster for final compression and some normalization on the entire mix. i usualy use a final limiter in the final mastering process, once all songs in a colection are complete... i am sure it can be used as you suggest.
