The difference is that ASIO2 allows direct monitoring from within a seq. app, thus is used always in xtc mode.
I personally use sfp mode and mix in scope, so I don't need it, and use Asio1, a.f.a.i.k. there should be no difference soundwise.

Both.
The fact is that audio tracks in the seq. are going throug it's mixer, and if routed out to a stereo asio they are also mixed/summed there, and these two processes affect the sound. Then there is the part in Scope, that for my ears is at an higher quality of a lot of native apps.
Having many dsp's, I prefer to route all tracks on separate busses in Cubase, each one to a channel of Asio, and effecting and mixing directly in Scope. Maybe it's just a matter of taste...but I like it much more.

ASIO2 also has a clock input. This makes for a sample tight like between VDAT for example, and your sequencer. Connect ASIO2 Dest Clock input to the Clock out of VDAT or VRC, and set sequencer to ASIO Positioning Protocol. Set same framerate, and both VDAT and sequencer will play sync, controlled by VRC.
